Anlegget har ett slusekammer med løftehøyde på 5 meter og et flomslusekammer. Nedenfor sluseanlegget er den en ventebrygge for fritidsbåter.
Skien sluse ligger sentralt plassert i byen, i vannskillet mellom Hjellevannet og Bryggevannet, mellom Skien Mølle mot nord og Smiøya mot sør. På Smiøya ligger den fredete fabrikkbygningen PM5 (papirmaskin nr. 5) fra 1882.
Sluseanlegget er modernisert i flere omganger. Den manuelle manøvreringen ble automatisert allerede i 1909. Sluseportene av tre ble først erstattet med jernrammer med tredekke og siden med stålporter i 1980-åra, mens flomporten fremdeles har jernramme med tredekke. Sluseveggene er fuget og støpt ut for å lette slusing av tømmer. På oversiden av slusa mot Hjellevannet er det bygd ny hydraulisk bruklaff i stål med betongdekke. I 2004 ble sluseanlegget lyssatt som en del av lysprosjektet ”Morild”.
